The W. M. Keck division of the CLAS Bioimaging Facility supports a broad range of individual and collaborative research by over 150 scientists in the Phoenix metro area and throughout the state. It is managed by Dr. D. Page Baluch and is housed within the School of Life Sciences at ASU's Tempe campus. In addition to providing light microscopy and related resources to researchers, the Keck lab participates in the BIO504 and 503 Bioimaging courses at ASU, conducts training and outreach events and collaborates with university partners such as Barrow Neurological Institute, Mayo Clinic Scottsdale, USDA, Midwestern University, University of Arizona, Maricopa Community College and NAU. The core lab houses some of the latest bioimaging technology including; laser scanning confocal, light sheet, fluorescence, live cell, and multi-photon microscopy. In addition to microscopy, the core also has an  in-vivo small animal imaging system, a nano-injection system, cryo- and paraffin embedded histology equipment and supporting cell culture resources for live cell imaging.

Biosafety Protocols for Live Cell Imaging

Thumb Live sample imaging is becoming a routine method of imaging cells but require that additional precautions be established to ensure a safe environment within a shared facility. This link contains the guidelines established by the Arizona State University EHS and Keck Bioimaging Laboratory. These define the cell lines and organisms permitted in the Keck lab and the procedures for cleanup and disposal of contaminated materials.

Evos FL Auto Live Cell Imaging System

Thumb The Evos inverted microscope system is equipped with an onstage incubator for environmental control and can accommodate multi-well plates and standard round culture dishes. The fluorescent/widefield system is capable of z stage movement and tiling. Phase corrected objectives include 4xPH, 10xPH, 20xL PH, 40xL PH as well as a 60x 1.42NA oil lens for higher resolution imaging. Four LED light cubes are available to cover the standard DAPI, GFP, RFP and Cy5 wavelengths.

Leica CM1950 Cryostat

Thumb Leica CM1950 cryostat has a precise microtome object feed mechanism for high-quality sections, CryoZone™ system for optimal sectioning conditions and UVC disinfection and AgProtect™ for operator safety.

Leica RM2235 Microtome

Thumb Designed for manual routine paraffin embedded tissue sectioning.

Leica SP8 White Light Laser Confocal/Lightsheet

Thumb The Leica SP8 White Light Laser Confocal/Lightsheet inverted microscope system is designed for confocal live and fixed cell imaging. It is equipped with a white light tunable laser as well as Argon [488] and 405nm lasers. The system has a combination of PMT and HyD GaAsP detectors as well as a DIC bightfield detector. The system allows for standard confocal imaging with or without additional resonant scanning speed or can be switched into live cell mode by adding the Tokai Hit stage top incubation chamber and/or Light Sheet module and objectives.

Leica TCS SP5 AOBS Spectral Confocal System

Thumb The Leica SP5 has four laser lines (405, 488 [458, 476, 496,514], 561 and 633) and is capable of capturing 5 channels which includes a DIC bright field detector. The microscope is an upright DM6000 and has a motorized stage. Other features include; pixel resolution up to 8000x8000, AOTF laser modulation, 200 degree field rotation, standard objective set plus a 20x multi-emersion objective, electrophysiology software package and a Bioptics environmental chamber unit.

Nikon Elipse TE300 Inverted Video Microscope

Thumb This imaging system consists of a Nikon Elipse TE300 inverted microscope and is outfitted with two cameras; a Hamamatsu Orca 3CCD and a Olympus DP26 color camera that allows real-time digital image capture. Both fluorescent and visible light (including both Phase and DIC contrast enhancement capabilities) are supported.

Nikon Microscope w/ Nanoject III

Thumb Nikon microscope equipped with Eppendorf micromanipulation controllers and a Drummond Scientific Nanojet III. Automated microprocessor controlled Microinjection Pipet delivering precise nanoliter volumes accurately. Remote control and non-rotating plunger eliminates vibration allowing precise injection minimizing possible cell rupture. Selectable Injection Volumes: from 2.3 nanoliters to 69.0 nanoliters in 16 increments.

Prairie Ultima IV 2-Photon Microscope

Thumb The Prairie Ultima IV 2P microscope comes equipped with an Ultima IV in vivo laser scanning microscope with an X-Y translation stage. The dual laser system includes the Mai Tai and Tsunami lasers capable of MPE imaging, oblation and uncaging.

Technical Assistance

Thumb Training to use the instruments within the Keck Bioimaging Laboratory is free of charge. A technical assistance fee is charged for any work or imaging that is done as a service for a lab.
